Today was busy day for me, I got the reply from my supervisor concerning my project. I have to revise paper. I have to add references, tables, graphics, etc..and etc..hmm I was so tired today…Going home at the end of the day, I though there were some foods at home….yeah I got the foods…sorry, I could not add any comments I was sleepy …..zzzzzzzzzzzz…
Melihat berita tentang tes keayahan atau peternity test yang selama ini semakin sering terdengar, jadi pengin sharing tentang bagaimana cara menentukannya..OK di postingan ini akan saya sampaikan sebenarnya bagaimana DNA itu bisa mengurai siapa ayah kita, atau siapa ibu biologis kita yang sebenarnya…
Kita sudah tahu bahwa setiap sell di tubuh kita adalah identik, artinya untaian DNA dari rambut sampai sel darah putih adalah sama. DNA snediri terdiri atas 4 basa A,C,G, dan T yang beruntun membentuk untaian panjang. Setiap DNA pada individu adalah unik, setiap individu tidak sama DNA nya dengan DNA orang lain, Kecuali orang kembar identik (yang berasal dari satu telur dan satu seprma). Unik di maksudkan adalah pada dua orang jika di perhatkan akan adanya variasi perbedaan basa pada setiap seribu basa atau lebih, jika disebut maka ada istilah Polimorfisme. Inilah secara statitik DNA setiap orang 99,99 % adalah berbeda.
DNA kita terdiri atas sumbangan DNA bapak dan ibu kita. DNA kita 99,99 % adalah sama dengan orang tua kita. 50 % dari bapak, 50% dari ibu.DNA kita juga mirip dengan DNA kakek nenek kita, juga ada bagian yang sama dengan sepupu atau keponakan kita. Nah Paternity test menggunakan figure DNA yang sama untuk memnentukan sipa orang tua kita sebenarnya.
DNA fingerprinting digunakan untuk menyelidiki kasus ini. Teknik ini biasanya menggunakan enzym restriksi yang spesifik akan memotong untainan DNA pada titik tertentu yang dikehendaki. Setiap enzym mempunyai tempat pemotongan tertentu.
Sebagai contoh, EcoRI memotong DNA pada untaian GAATTC dan akan hanya memotong tempat tempat yang ada untaian ini.

Tekniknya adalah, DNA dari si anak, dari si ibu, dan dari 2 orang yang dicurigai sebagai ayahnya di tes dengan teknik ini. kemudian hasilnya dibandingkan. Pola DNA dari hasil teknik ini kemudian di cocokan untuk mencari tinghkat kesamaan dari masing -masing DNA.
Sebagai contoh pada gamabr di samping EcoR1 memotong DNA mary hanya pada 1 tempat, sedangkan memotong dua orang yang mengaku ayahnya ) (Bob dan Larry pada tempat yeng berbeda.
Kemudian DNA si anak di cocokan dengan ketiga sample di atas. Kemudian didapatkan hasil yang sedemikian sehingga dpat diketahui kecocokan si anak dan si ayah.
Kemudian bagaimana kedua ayah ini memiliki DNA yang mirip, dan pemotongannya sama dengan si anak, bagaimana cara menentukannya ? kita tinggal menambah tempat pemotongan dengan enzym lain dan mencari kesamaan hasilnya seperti tersebut di atas. maka kita dapat pola yang benar-benar ada perbedaan antara kedua supsect ayah tersebut…

You should have been familiar with the term of “DNA“. But did you know about the function of this material ?. Of course most of you also already knew that. This section, hopefully, could inform you about the process how the DNA will be used for analysis.
1. DNA extraction : this is first step. DNA is isolated from the cells (any nucleated cells in the body can be used). Usually scientists used leukocytes in the blood to get enough DNA. Some forensic studies used material from sperm, hairs, and saliva as well. You should know it at the previous posting. This is the process in the chart.
2. After extracting DNA,Scientist will use it for many necessities, such as PCR, MLPA, Shoutern blotting, DGEE, and an others. The most technic using by is PCR aka Polimerase Chain Reaction. You can find this machine in the picture. You see that the machine is not big enough. its like a box in your home. The tube used is also smaller than your pen.
3. After several reaction, you can see it in HERE, they will get the product called as PCR product. Than they will load the product in the gel,usually Agarose gel or Poliacrilamide gel to show the presence of DNA interested. We than visualize the DNA by make a photo in the UV light (of course with some treatment). The DNA will be shown as bands in the gel.

Pagi ini bener-bener aku gak bisa bangun fresh, kepala pusing, mah…mata berkunang-kunang, jalan juga sempoyongan…waduh bener-benar dah…eit tapi bukan sedang mabuk ya, atau abis minum wisky…(seperti kebiasaan orang-orang sini)…
Malam tadi bener-bener gak bisa nyenyak tidur….banyak pikiran yang harus aku pikirkan….belum lagi ada “tambahan’ problem yang tidak aku agendakan……
Satu yang jadi ganjalan adalah aku lupa kalo kemaren anakku genep 4 tahun…waduh..mana pula ini…!!, bener-bener kelupaan…mungkin karena tugas yang banyak apa ya…tapi gak juga deh kayaknya… BTW itu yang membuat aku gak bisa tidur…sedih rasanya gak bisa “pas” menyampaikan ….”Kakah, sekarang adalah 4 tahun hari lahirmu, tambah pinter ya, tambah nurut, baik pada orang tua, saudara, dan temen2…etc,etc…
Yah untungnya pagi bisa nyambung telepon jadi sedikit mengobati rasa kesalahanku…”maaf ya Kak..”

OK today I will give you ” a teaching “. The fact you can do it in your home with your equipments, even in your kitchen…Are you ready ? OK here they are. You can simply can collect your DNA ( yeah its really…. your own DNA)…and stored in your own freezer …What for…?? Aha…you can use it just for your collection or may be you want to know about the probability whether you will get some disease in the future…..Or Paternity test for your kid….??? ha…ha… just a joke…!!

You must have a question concerning about it…let me explain it…
1) Where did the DNA I am seeing come from?
You have DNA in every cell in your bodies but this DNA comes from the components of your saliva: epithelial (or cheek lining) cells and B-cells (immune cells of the same kind found in tears, saliva, and sweat).
2) Chemically, how does this experiment work?
The principle of our DNA extraction is simple: break open the cells with soap (which solubilizes them ( Drew, do I need to explain this and if so how?), and precipitate the DNA out of its usual state in solution using alcohol (that is, the alcohol is essentially pulling the water out of the DNA).
3) But is this ‘real science’?
DNA extraction procedures in ‘real world’ situation (crime labs, genetic testing, etc.) follow basically the same principles that we did – with some variations and (obvious!) refinements. For example, more DNA can be obtained using a refined human sample – cheek cell swab, rather than saliva — and another way to break the cells open is to add gentler detergents and enzymes, to keep the DNA intact without attached proteins.
4) Can I do this at home?
You could try but with any other bodily tissue or fluid, the mixture would require more incubation for cells to break down – and it precipitation will not occur without 100% (200 proof) pure alcohol.

Yesterday afternoon, I compiled several resources from my computer to be presented today. This is about our life, our origin. You may be already know about it. But I think it is great to show you, how our life becomes so beautiful like now.
1. our life is began with small material, sperm and eggs.
2. cells developed. After the fertilization the cells will develop, we usually than called as morula, blastula, gastrula, and then embryo.
3. organs developed. After that you will see amazing development. your head, hand, leg, and all of the organ in our body will be developed continuously, and rapidly.
People, I show you this chapter as now most of the people in the world did not know where they origin actually came from. This video just remind us that all of us came from same materials, small thing, people usually don’t care about it, Sperm and Egg. May we conscious about this fact. So the war, racialism, conflict will be disappear from the world. let’s hope..

Today, I will give you a “teaching”, may be it will help you to prevent this disease so early. Do you know that this disease has be the number one of the most perfect killer in the world ? OK, let’s begin. Coronary heart disease (CHD), also called coronary artery disease usually develop when a combination of fatty material, calcium, and scar tissue (plaque) builds up in the arteries that supply the heart with blood. Through these arteries, called the coronary arteries, the heart muscle (myocardium) gets the oxygen and other nutrients it needs to pump blood.The plaque often narrows the artery so that the heart does not get enough blood.
Hey man you have to be careful if you have chest pain, we called as angina. If plaque completely blocks blood flow, it may cause a heart attack (myocardial infarction) or a fatal rhythm disturbance (sudden cardiac arrest).
The heart consists of 4 chambers: an atrium and a ventricle on the right, and an atrium and ventricle on the left. Blood returning to the heart from veins all over the body flows into the right atrium. From there the blood flows into the right ventricle, which pumps it out to the lungs for oxygenation.The oxygen-rich blood returns to the left atrium. From there the blood flows into the left ventricle, which pumps it at high pressure into the arteries. This entire process constitutes one heartbeat.

The pumping, or contraction, of the left ventricle must be very powerful because that is what keeps the blood flowing throughout the body. The strength of the heart muscle depends on the oxygen and nutrient supply coming via the coronary arteries. These arteries are usually strong, elastic, and quite flexible.The heart has 3 major coronary arteries. Two of these arteries arise from a common stem, called the left main coronary artery.The left main coronary artery supplies the left side of the heart. Its left anterior descending (LAD) branch supplies the front part of the heart. The left circumflex (LCX) branch supplies the left lateral and back side of the heart. Finally, the right coronary artery (RCA) is separate and supplies the right and the bottom parts of the heart.
As a child, the inner lining of the coronary arteries is quite smooth, allowing blood to flow easily. As a person ages, the cholesterol ( hey friend you have to prevent this stuff )and calcium content in the walls of the coronary arteries increases, making them thicker and less elastic. Unhealthy habits, such as a diet high in cholesterol and other fats, smoking, and lack of exercise accelerate the deposit of fat and calcium within the inner lining of coronary arteries.
This process is known as atherosclerosis, or hardening of the arteries.
The deposits, or plaques, eventually obstruct the blood vessel, which begins to restrict blood flow. Plaque is like a firm shell with a soft inner core containing cholesterol. As blood hits it during each heartbeat, the plaque may crack open and expose its inner cholesterol core, which promotes blood clotting. Clots may further reduce blood flow, causing severe pain (angina), or even block it all together.

Jam menunjukan pukul 08.30 pagi, aku pikir semuanya udah aku siapkan.. buku lab udah aku masukkan tas, kertas, bulpen, kunci locker ( uhh hampir aja lupa..), udara di luar kayaknya dingin banget nih…jaket tidak lupa aku siapkan…hmmm…terasa udah semuanya masuk…apa ya yang lupa…masih ku pikir-pikir…kayaknya masih ada yang ‘hilang’ ini, sesuatu yang rutin tapi aku lupa apa ??…otakku mulai berputar-putar…Oh…ternyata….”mandi”…!! dasar…
Selesai mandi tas ku siapkan di meja, jaket, sepatu…semuanya kayaknya udah sip nih…eit–eit ..ternyata masih ada yang kurang…bekal makan belum aku siapkan…harus cepat-cepat ni..mana jam udah siang lagi…..setiap Senin pagi jam 9 ada work discusion , dan kali ini temanya adalah Fragile X…waha kudu ikut nih…Aku sambar aja nasi kemaren, tempe, dan ikan teri…hmm cukuplah aku pikir…Sip-sip..semuanya dah siap…Eng ing eng….sepeda aku kayuh…wah ternyata…cuaca tidak bersahabat nih…angin nya kenceng banget…uh pengin rasanya segera sampai di Maastunel…
Singkat cerita udah sampai nih…di laboratorium. Suasananya kok sepi , gak kaya biasanya..temen-temen di lab juga kok nongol-nongol…pada kemana nih aku pikir-pikir…meja samping gak ada peralatan, mesin PCR pada nganggur….microtom tergeletak begitu saja…akh mungkin belum pada datang pikirku….tapi kok ditunggu-tunggu gak muncul-muncul….
Hmmm ternyata oh ternyata…hari ini adalah hari libur lab…..yah jadinya…sendiri di lab. utak utik sana sini ampe jam 12…akhirnya go home….hhmm hari yang aneh …..!!
