A University of California Riverside team of scientists has found that an available drug called minocycline, used widely to treat acne and skin infections, might also be used to treat Fragile X.

The study’s findings have already led to the approval of a FRAXA-funded clinical trial in Toronto, Canada, that will test minocycline in patients with Fragile X.

Neurons in the brain communicate with each other at specialized contact sites called synapses, with many of these synapses occurring on small mushroom-shaped structures called dendritic spines.

During early development dendritic spines have immature finger-like shapes. But learning stabilizes the synapses and dendritic spines take on a mature mushroom shape, which make them more efficient.

The brains of patients with Fragile X syndrome have an overabundance of immature dendritic spines.

In their report, the researchers, led by Iryna Ethell and Douglas Ethell, faculty members in UCR’s Division of Biomedical Sciences, describe how dendritic spine development in mice with Fragile X is delayed by enzymes called matrix metalloproteinases (MMPs), which are involved in normal brain development and physiological processes. They report that high levels of certain MMPs keep the synapses immature and inefficient.

But minocycline, they found, reduces these MMP levels in the mice, allowing the synapses to mature and make more efficient contacts between neurons in the brain. The outcome: corrected brain abnormalities in dendritic spines, reduced anxiety and improved cognitive function.

Study results appear online, ahead of print, in the Journal of Medical Genetics.

In their experiments, the Ethells found that young Fragile X mice treated with minocycline showed an increase of dendritic spine maturation in the hippocampus, a brain area that is critical for learning and memory. Besides less anxiety, minocycline-treated mice showed better exploration skills as compared to untreated mice.

The Ethells are enthusiastic about how their discovery already is leading to a clinical trial.

“Clinical studies often quickly follow such basic science because once there is a solid understanding of how problems arise, it is much easier to come up with solutions,” said Iryna Ethell, an associate professor of biomedical sciences.

The study was funded by a grant from FRAXA.

Dr. Michael Tranfaglia, FRAXA’s chief scientific officer, said of the UCR researchers, “This group has done something unique and incredibly valuable: They have identified an off-the-shelf treatment for Fragile X through their basic research. By bringing their unique perspective to Fragile X research, they have helped us to understand why neurons are malformed in this disorder, and more importantly, how we can treat it.

“We were so impressed with their work that we just awarded Dr. Iryna Ethell the FRAXA Breakthrough Award for 2008. This is easily the most important scientific breakthrough in the Fragile X field in many years.”

According to Dr. Carlo Paribello, president of Fragile X Research Foundation of Canada and the director of the clinical trial (scheduled for early 2009) at Surrey Place Centre Fragile X Clinic in Toronto, Canada, the UCR-led study “will go a long way towards dispelling the idea that mental impairment cannot be treated.”

UCR’s Douglas Ethell, an assistant professor of biomedical sciences, noted that effective therapies for Fragile X syndrome are few and far between. “This is a good time for identifying highly effective therapeutic strategies that might work in Fragile X patients,” he said. “We are excited that our research has the potential to affect many lives.”

Minocycline belongs to a group of antibiotics that has been used in people for more than fifty years to treat Lyme disease, acne, and other skin infections.

Minocycline may have beneficial effects in other disorders where higher-than-normal brain levels of MMP-9 are found. It is currently under study for treating rheumatoid arthritis, multiple sclerosis (MS), Parkinson’s disease, and several other neurodegenerative conditions.

“In the future, new compounds that more specifically target MMP-9 can be developed and tested,” Douglas Ethell said.

Next in their research, the Ethells and their colleagues plan to refine the therapeutic strategy in Fragile X mice to determine the optimal age, if any, to administer minocycline. They will also explore other MMP inhibitors that may be more effective than minocycline.

“We will investigate whether a combination of MMP inhibitors with other drugs, such as fenobam, can help mature the synapses in Fragile X mice,” Iryna Ethell said.

UCR’s Office of Technology Commercialization has applied for a patent on the discovery by the Ethells and their collaborators, with an interest in finding partners to accelerate development of treatments for Fragile X syndrome and other forms of mental retardation and autism.

Melihat berita tentang tes keayahan atau peternity test yang selama ini semakin sering terdengar, jadi pengin sharing tentang bagaimana cara menentukannya..OK di postingan ini akan saya sampaikan sebenarnya bagaimana DNA itu bisa mengurai siapa ayah kita, atau siapa ibu biologis kita yang sebenarnya…

Kita sudah tahu bahwa setiap sell di tubuh kita adalah identik, artinya untaian DNA dari rambut sampai sel darah putih adalah sama. DNA snediri terdiri atas 4 basa A,C,G, dan T yang beruntun membentuk untaian panjang. Setiap DNA pada individu adalah unik, setiap individu tidak sama DNA nya dengan DNA orang lain, Kecuali orang kembar identik (yang berasal dari satu telur dan satu seprma). Unik di maksudkan adalah pada dua orang jika di perhatkan akan adanya variasi perbedaan basa pada setiap seribu basa atau lebih, jika disebut maka ada istilah Polimorfisme. Inilah secara statitik DNA setiap orang 99,99 % adalah berbeda.

DNA kita terdiri atas sumbangan DNA bapak dan ibu kita. DNA kita 99,99 % adalah sama dengan orang tua kita. 50 % dari bapak, 50% dari ibu.DNA kita juga mirip dengan DNA kakek nenek kita, juga ada bagian yang sama dengan sepupu atau keponakan kita. Nah Paternity test menggunakan figure DNA yang sama untuk memnentukan sipa orang tua kita sebenarnya.

DNA fingerprinting digunakan untuk menyelidiki kasus ini. Teknik ini biasanya menggunakan enzym restriksi yang spesifik akan memotong untainan DNA pada titik tertentu yang dikehendaki. Setiap enzym mempunyai tempat pemotongan tertentu.

Sebagai contoh,  EcoRI memotong DNA pada untaian  GAATTC dan akan hanya memotong tempat tempat yang ada untaian ini.

Tekniknya adalah, DNA dari si anak, dari si ibu, dan dari 2 orang yang dicurigai sebagai ayahnya di tes dengan teknik ini. kemudian hasilnya dibandingkan. Pola DNA dari hasil teknik ini kemudian di cocokan untuk mencari tinghkat kesamaan dari masing -masing DNA.

Sebagai contoh pada gamabr di samping EcoR1 memotong DNA mary hanya pada 1 tempat, sedangkan memotong dua orang yang mengaku ayahnya ) (Bob dan Larry pada tempat yeng berbeda.

Kemudian DNA si anak di cocokan dengan ketiga sample di atas. Kemudian didapatkan hasil yang sedemikian sehingga dpat diketahui kecocokan si anak dan si ayah.

Kemudian bagaimana kedua ayah ini memiliki DNA yang mirip, dan pemotongannya sama dengan si anak, bagaimana cara menentukannya ? kita tinggal menambah tempat pemotongan dengan enzym lain dan mencari kesamaan hasilnya seperti tersebut di atas. maka kita dapat pola yang benar-benar ada perbedaan antara kedua supsect ayah tersebut…

You should have been familiar with the term of “DNA“. But did you know about the function of this material ?. Of course most of you also already knew that. This section, hopefully, could inform you about the process how the DNA will be used for analysis.
1. DNA extraction : this is first step. DNA is isolated from the cells (any nucleated cells in the body can be used). Usually scientists used leukocytes in the blood to get enough DNA. Some forensic studies used material from sperm, hairs, and saliva as well. You should know it at the previous posting. This is the process in the chart.

2. After extracting DNA,Scientist will use it for many necessities, such as PCR, MLPA, Shoutern blotting, DGEE, and an others. The most technic using by is PCR aka Polimerase Chain Reaction. You can find this machine in the picture. You see that the machine is not big enough. its like a box in your home. The tube used is also smaller than your pen.

3. After several reaction, you can see it in HERE, they will get the product called as PCR product. Than they will load the product in the gel,usually Agarose gel or Poliacrilamide gel to show the presence of DNA interested. We than visualize the DNA by  make a photo in the UV light (of course with some treatment). The DNA will be shown as bands in the gel.

Pagi ini bener-bener aku gak bisa bangun fresh, kepala pusing, mah…mata berkunang-kunang, jalan juga sempoyongan…waduh bener-benar dah…eit tapi bukan sedang mabuk ya, atau abis minum wisky…(seperti kebiasaan orang-orang sini)…

Malam tadi bener-bener gak bisa nyenyak tidur….banyak pikiran yang harus aku pikirkan….belum lagi ada “tambahan’ problem yang tidak aku agendakan……

Satu yang jadi ganjalan adalah aku lupa kalo kemaren anakku genep 4 tahun…waduh..mana pula ini…!!, bener-bener kelupaan…mungkin karena tugas yang banyak apa ya…tapi gak juga deh kayaknya…  BTW itu yang membuat aku gak bisa tidur…sedih rasanya gak bisa “pas” menyampaikan ….”Kakah, sekarang adalah 4 tahun hari lahirmu, tambah pinter ya, tambah nurut, baik pada orang tua, saudara, dan temen2…etc,etc…

Yah untungnya pagi bisa nyambung telepon jadi sedikit mengobati rasa kesalahanku…”maaf ya Kak..”
